Objectives Chemotherapy-induced peripheral neuropathy (CIPN) is a major factor contributing to decreased quality of life in patients with cancer undergoing or having completed chemotherapy. This study evaluated the efficacy and safety of mirogabalin in the treatment of CIPN.Methods In this single-centre retrospective study, 100 patients were enrolled consecutively with advanced cancer who received mirogabalin for CIPN between June 2019 and December 2023. The efficacy of mirogabalin was assessed using the Eastern Cooperative Oncology Group (ECOG) neurosensory score, and safety was evaluated using the Common Terminology Criteria for Adverse Events.Results 13 patients were excluded because of missing data or loss to follow-up, resulting in 87 patients in the final analysis. The proportion of patients whose ECOG neurosensory score improved from baseline was 26.4% at week 4 (p<0.01) and 36.8% at week 8 (p<0.01). Mirogabalin significantly ameliorated CIPN at both week 4 and 8 (both p<0.01) regardless of whether chemotherapy was ongoing or completed. In patients treated with an effective dose of mirogabalin, CIPN was significantly improved at both week 4 and 8 from baseline (both p<0.01). Treatment-related adverse events were observed in 29 patients (33.3%). Among patients receiving ongoing chemotherapy, dose reduction because of CIPN was required in two patients (3.6%) and treatment discontinuation occurred in one patient (1.8%).Conclusions Treatment with an effective dose of mirogabalin suggests an improvement in CIPN induced by various neurotoxic chemotherapeutic agents regardless of whether chemotherapy was ongoing or completed.
