Introduction Calcinosis cutis (CC) is a painful deposition of calciumphosphate in the skin, frequently observed in systemic sclerosis (SSc) and dermatomyositis (DM). CC contributes substantially to morbidity and impaired quality of life, yet no standard therapy currently exists. So far, no systematic studies have addressed how to best characterize the structural or compositional features of CC lesions.Material and Methods As part of the multicenter study CURING CALCINOSIS, ex vivo CC lesions are collected from patients with SSc and DM. Using a combination of complementary methods, including advanced imaging techniques, we aim to describe structural and compositional variation in CC lesions.Results Pilot analyses indicate that the applied approaches are feasible for systematic evaluation of CC architecture and composition.Conclusions The project aims to provide the first systematic ex vivo characterization of the structural and compositional variation in CC lesions from patients with SSc and DM. These insights may help identify CC lesion characteristics that could guide and improve clinical treatment strategies.Abstract P.222 Figure 1
