Objectives The primary objective of EAFToS part I is to characterize the onboarding experience and utilization of OMB in patients with RMS in Australia. Secondary objectives are to describe the profile of patients initiating OMB, evaluate patient demographics and prior therapy.Methods The primary endpoint was the proportion of doses not completed within 3 days of the expected date during initiation and the first 3 months of maintenance. Additionally, treatment adherence, compliance and persistence were measured.Results The interim analysis (data cutoff: Aug 19, 2024) included 662 de-identified patients initiated on MSGo who were predominantly aged 41–50 years (29%). Most patients were female (75%), and received prior therapy (73%). Most patients were adherent within the expected timeframe (proportion of patients with no non-adherent dose: 0.88 [95% CI: 0.85–0.90]). High persistence rates to treatment were observed at 91 days (proportion: 0.926 [0.903–0.944]), 365 days (0.820 [0.786–0.850]), and 730 days (0.737 [0.694–0.774]) after the first dose. Prior treatment and age >60 years (p<0.001 for both) were associated with shorter time on OMB. Compliance to OMB was at least 80% with 96.2% (634/659) patients and at least 90% with 91.7% (604/659) patients.Conclusion High adherence rates during initiation and the first 3 months of maintenance, persistence beyond 91 days, and a high proportion of patient compliance were observed.
