1286 Developing patient-derived tumor organoid model for drug test in Vietnamese setting

Background Patient-derived tumor organoid (PDTO) represents a clinically relevant model for investigating and predicting therapeutic responses. Vietnam represents a critical yet underexplored setting for organoid-based research and applications.Methods Data are generated using PDTO-based drug sensitivity and clinical input.Results We report a colorectal cancer (CRC) case where organoid-based drug sensitivity correlated with clinical outcomes ( figure 1). The PDTOs are sensitive to 5-FU, oxaliplatin, irinotecan and their combination (figure 2). However, the 75-year-old male Vietnamese patient with liver-metastatic CRC harboring KRAS, PIK3CA, PTEN, and TP53 mutations exhibited differential treatment responses (figure 3):First-line therapy FOLFOX: tumor progression in metastatic lesions.Second-line therapy FOLFIRI: Significant tumor regression after 3 cycles.Conclusions A major limitation of this study is genetic profile of the established PDTO is not known and under investigation.PDTOs established from primary tumors and tumor profiling may reflect clinical response to chemotherapy in CRC patients with liver metastases.We aim to co-culture PDTOs with immune cells to leverage the platform for predicting tumor sensitivity to immunotherapy in Vietnam.Abstract 1286 Figures 1–3
